左心引流对离体大鼠心肌影响

摘要 目的本实验的目的是研究左心引流的心肌保护作用。方法将健康 SD 大鼠随机分为两组:左心引流组(A组)和对照组(B 组,不建立左心引流)建立改良式 Langendorff 离体鼠心灌注模型,将大鼠离体心脏置于模型中,A 组做相当于左心引流效应的干预,对 B 组则不进行任何干预处理。记录干预前、恢复后5、10、20、30、60分钟测定心功能指标(LVEDP、LVESP、LVDP、dP/dt),CK-MB 含量,全程心电活动以及左室心内膜下心肌组织送电镜检查。结果左心引流组和对照组在心肌舒张收缩功能、心肌酶 CK-MB 的指标具有显著性差异(P<0.05),心肌的电镜超微结构也有明显的不同。结论左心引流对心肌有更好的保护作用(P<0.05)。 Objective The purpose of this study is to assess the myoprotective effects of venting of left ventricle on Langendorff- perfused rat hearts. Methods There were two anesthetized S - D rat groups in this study named group A (n = 8, venting of left ventricle) and group B ( n = 8, control) . All samples' hearts were switched into improved Langendorff mode. Observed indexes include: myocardial contractility, ECG, myocardial CK - MB in blood and myocardial ultra structure observation from electron microscope. Results Venting group improved myocardial contractility and relaxation ( + dp/dtmax) as compared with control group during ischemic and repeffusion period (P〈 0.05). CK- MB leakage was reduced significantly in group A (P 〈 0.01 ). Myocardial electron microscope observation showed that myocardial ultra structure of group A was better than that of group B. Conclusion This experiment snggests that venting of left ventricule has cardio - protective effect and attenuates ischemic- repeffusion injury and necrosis distinctly even in the normal peffused beat hearts.
