鼻咽癌癌变过程中EBERs表达状况的研究

STUDY ON THE EXPRESSION OF EBERs IN NPC TUMORIGENESIS

本文采用重组质粒ｐＳＰ６５（带有ＥＢＥＲｓ的基因）体外转录合成的地高辛标记的单链反义ＲＮＡ探针，用原位杂交方法检测处于癌变不同阶段的鼻咽活检组织中ＥＢＥＲｓ的表达状况。ＥＢＥＲｓ检出率为：鼻咽癌（２３／２４），正常鼻咽组织，慢性炎症增生及单纯性增生和化生（０／３０），异型增生和化生（５／８），头颈部其它肿瘤（０／５），鼻咽癌颈淋巴结转移灶（１／１）。进一步证明：（１）ＥＢＶ与ＮＰＣ密切相关；（２）ＥＢＶ在ＮＰＣ转移过程中并不丢失；（３）ＥＢＶ可能并不感染正常鼻咽上皮。

In this study,EBERs specific ssRNA probes transcribed from the recombinant plasmidPSP65 in which the gens of EBERs were inserted were used to detect the expression of EBERs in nasopha-ryngeal biopsies(carcinoma and precancerous lesions)and other tumors of head and neck by in situ hy-bridization.As the results, 23 of 24 cases of NPC biopsies were positive to the EBERs anti-sense probes.EBERs were not detected in 20 cases of simple hyperplasia or metaplasia of nasopharyngeal mucosa and in 5cases of carcinoma of head and neck.One case of cervical lymphonode metastasis of NPC was EBERs posi-tive.Generally,the heteroplasia and heterometaplasia of nasopharyngeal mucosa are the precancerous lesionsof NPC.In these lesions,the positive rate of EBERs expression was 3/8.Our results suggest:(1) EBV wasclosely related to NPC;(2)EBV may not lost during NPC metastasis.(3)Normal nasopharyngeal epithelialcells may not be infected by EBV.
