摘要
建立了HPLC法测定人血浆中非诺贝酸。以苄普地尔为内标,采用C18柱,流动相为甲醇-水-10%磷酸(70∶30∶1),检测波长286nm。非诺贝酸在0.1~25μg/ml浓度范围内线性关系良好,方法回收率96.8%~105.0%。24名男性志愿者单剂量口服非诺贝特200mg的药物动力学参数Tmax、Cmax、t1/2和AUC0→∞分别为(6.02±2.56)h、(5.33±3.81)μg/ml、(23.61±5.98)h和(169.75±126.96)μg·h·ml-1。
A HPLC method for determination of fenofibric acid in human plasma was established. Bepridil was used as the internal standard. A C18column was used with the mobile phase of methanol-water-10% phosphoric acid (70: 30:1), at the detection wavelength of 286nm. The calibration curve was linear in the range of. 0.1 - 25μ/ml. The method recoveries were between 96.8 % - 105.0 %. The pharmacokinetics were investigated in 24 male volunteers with single dose of 200mg fenofibrate. The Tmax, Cmax, t1/2 and AUCo→∞were (6.02±2.56) h, (5.33 ±3.81) μg/ml, (23.61±5.98) h and (169.75 ± 126.96)μg·h·ml^-1 respectively.
