格列齐特对糖尿病大鼠心肌ATP敏感性钾通道mRNA表达的影响 被引量:1

摘要 目的:观察格列齐特(Gliclazide)对糖尿病大鼠心肌组织ATP敏感性钾通道mRNA表达的影响.方法:采用链脲佐菌素小剂量ip结合高脂饮食饲养形成2型糖尿病大鼠模型,随机分为糖尿病对照组、糖尿病胰岛素治疗组和糖尿病格列齐特组,另设正常对照组.RT-PCR检测ATP敏感性钾通道mRNA表达.结果:糖尿病组、糖尿病胰岛素治疗组和正常对照组ATP敏感性钾通道mRNA表达水平无差异(P>0.05).格列齐特治疗组ATP敏感性钾通道mRNA表达水平较正常对照组和糖尿病组无明显变化(P>0.05).结论:链脲佐菌素诱导的实验性糖尿病不影响心肌ATP敏感性钾通道mRNA表达水平;治疗剂量的格列齐特对ATP敏感性钾通道mRNA的表达无影响. AIM: To observe the effect of Gliclazide on mRNA expression of ATP-sensitive potassium channels(KATP) in myocardium of diabetic rats. METHODS: Rat diabetic model was made by streptozotozin ip and fed on high-fat diet for 6 weeks. The rats were then allocated randomly into 3 groups: the diabetes model group, the Gliclazide group and the positive control group (treated with insulin). A normal control group was also set up. KATP-mRNA expression in myocardial cells of the rats was detected by the reverse transcriptase-polymerase chain reaction (RTPCR). RESULTS: There was no significant difference in mRNA expression levels of KATP in heart between diabetic, insulin-treated diabetic and normal control groups ( P 〉 0.05 ). Comparison between Gliclazide-treated diabetic and non-treated diabetic rats showed that there was no change in mRNA levels of KATP in heart ( P 〉 0.05). CONCLUSION: The streptozotocin-induced diabetes itself does not affect the mRNA expression of KATP (SUR2 and Kir6.2) in myocardial tissues. Gliclazide with treatment dosage has no effect on gene expression of KATP.
