摘要
微卫星现为我国家禽分子生物学研究领域中的一大热点,在利用微卫星分析家禽遗传多样性时,其试验结果会受到采样、引物选择及数据分析等因素的影响。本文结合自身用30个微卫星对我国76个地方鸡种进行遗传多样性分析时的具体情况分别进行阐述。并指出要克服这些因素的影响,采样时尽量选择有品种代表性的群体,采样数达60羽,公母比1∶4;选择的引物应分散在不同的染色体上,引物的个数应不低于25对;在凝胶条带判型时最大限度的减少认为因素;数据分析应用规范、大家广泛使用认可的生物学软件。
Microsatellite is the hot point molecule biological research in china. Using microsatellite to analyse the genetic diversity of poultry, the results will be affected by the factors of sample collection, primers choosing,data analysis and so on. Contacted with the article in which the genetic diversity of seventy-six local chicken breeds in china were studied with thirty microsatellite markers,the particular things were expatiated separately. To overcome the factors' affection, in sample collection, the representive group in breed should be choosed as possible as, the sample must be achieved to sixty, the rate of male and female is one to four. The primers should spread around different chromosomes, the number of primers should be at least twenty-five pairs, Genotyping with gel bands, the man-made factor should be reduced as possible as, data analysis should be used by the standard and authorized bio-soft.
