白介素-18对树突状细胞瘤苗的抗肿瘤作用

摘要 目的:进一步提高树突状细胞(DC)瘤苗的抗肿瘤作用。方法:以腺病毒为载体转染目的基因至DC,研究DC瘤苗对荷瘤小鼠的免疫治疗作用。分别以乳酸脱氢酶(LDH)释放法、酶联免疫吸附试验(ELISA)检测细胞毒性T淋巴细胞(CTL)、自然杀伤细胞(NK)活性和细胞因子的分泌水平。结果:IL18/gp100DC瘤苗对荷瘤小鼠具有显著的治疗作用。IL18/gp100DC瘤苗治疗后,能明显增强CTL和NK活性,提高IFNγ和IL2的分泌水平,使体内肿瘤细胞发生明显坏死,瘤体及瘤周有大量炎性细胞浸润。CD4+T细胞和CD8+T细胞为发挥抗肿瘤作用所必须,NK细胞也起到一定的作用。结论:辅助性T细胞1(Th1)型细胞因子IL18基因修饰的gp100DC瘤苗能有效地诱导抗肿瘤免疫反应。
