摘要
目的建立多药耐药细胞系A549/ADM和初步探讨人肺腺癌细胞多药耐药的发生。方法对肺腺癌细胞A549采用持续低浓度和逐渐增加阿霉素浓度间歇诱导,建立多药耐药细胞系;使用流式细胞仪器分别检测敏感细胞系和耐药系的P-gp和MRP的表达;采用MTT法检测其对多种常用化疗药物的耐药程度。结果与母细胞系相比,耐药细胞系A549/ADM对ADM的耐药程度增加了14.29倍,对VCR、DDP、VP-16均有不同程度的耐药,对5-FU无耐药性;耐药细胞系A549/ADM表面P-GP、MRP表达分别为74.8%和61.2%(P<0.01)。结论A549/ADM对多种常用肿瘤化疗药物表现有不同程度的耐药,其耐药性可能与P-GR和MRP高表达有关。
Objective To establish a muhidrug -resistant cell line (A549/ADM) and investigate the mechanisms of muhidrug resistance in human lung adenocarcinoma cell line A549/ADM. Methods To establish a muhidrug - resistant cell line, A549 cells were cultured with continuous low - concentration and intermittent gradual - increasing - concentration of ADM. The expressions of P - GP and MRP were detected with flow cytometry. The resistance to chemotherapeutic drugs was evaluated with MTI' assay. Results A549/ADM cells were proved muhidrug - resistant. The resistance index of A549/ADM to ADM was 14.29 and the cells showed resistance to DDP,VCR, VP- 16 but not to 5 -FU; the expressions of P-gp and MRP enhanced significantly (74.8% and 61.2% respectively) compared to A549. Conclusion A549/ADM cells were resistant to clinically common used anti -cancer chemotherapeutic drugs and the resistance might related to P-gp and MRP overexpression.
