高效液相色谱法测定复方雷公藤药膜的含量

HPLC determination of triptolide in compound Leigongteng oral pellicles

目的：建立高效液相色谱法（HPLC）测定复方雷公藤药膜中雷公藤内酯醇的含量。方法：采用C18色谱柱，以甲醇-水（45：55）为流动相，检测波长218nm，进样量10μL。结果：线性范围为16～512ng，r=0．9999，平均回收率为98、8％，RSD为1．4％（n=6）。结论：HPLC法结果准确，重现性好，可用于测定药膜中雷公藤内酯醇含量。

OBJECTIVE To establish an HPLC method for the determination of triptolide in compound Leigongteng oral pellicles. METHODS DIANONSIL（^TM） C18 column（4.6 mm× 250 mm,5 μm）was used and the mobile phase consisted of methanolwater（45：55 ） at the flow rate of 1 mL·min^-1. The detection wavelength was 218 nm. The column temperature was 35 ℃. RESULTS The calibration curve was linear in the range 16 -512 ng （r = 0. 999 9）. The mean recovery was 99% （RSD = 1.4%, n =6 ）. CONCLUSION The method is sensitive. It can be used as a method of assay of the oral pellicles.
