甘蓝型油菜与芸芥属间杂种F_1的获得及鉴定 被引量:4

摘要 常规有性杂交很难获得油菜与芸芥的属间杂种。通过对早期杂种胚胎的子房培养,获得128株小苗。经形态学鉴定,其中24株为真杂种。运用分子标记技术(RAPD)进行真假杂种检测,87%的单株带有芸芥特异的标记带。在培养中,适宜的生长素和细胞分裂素浓度以及恰当的子房离体前的发育时间能提高成胚数和成胚率。 It is difficult to obtain the intergeneric hybrid between Brassica napus and Eruca sativa through the routine crossing. Ovary culture of hybrid embryos was used at early phase and 128 seedlings were obtained successfully. After morphological observations 24 seedlings were identified as potential hybrid. The analysis of random amplified polymorphic DNA (RAPD) showed 21 out of 24 F1 hybrid progenies (87 % ) carrying Eruca genome-specific RAPD markers, providing further evidences to support the speciation of these hybrids. Suitable hormone and cytokin concentration as well as time for excising and the pecentage of hybrid embryos ovary were crucial for increasing the number of hybrid embryos and the pecentage of hybrid embryos.
