17β-雌二醇对人骨肉瘤MG-63细胞株IL-6、IL-11和NF-κB表达的作用 被引量:3

摘要 目的观察17β-雌二醇(17β-E2)对人骨肉瘤(Osteosarcoma)MG-63细胞株IL-6(inter-leukin-6)、IL-11及NF-κB(nuclearfactor-kappaB)表达的影响。方法用RT-PCR技术、Northernblot和WesternBlot观察17β-E2对人骨肉瘤MG-63细胞株IL-6、IL-11及NF-κB表达的影响。结果17β-E2下调MG-63细胞IL-6mRNA及蛋白质的表达;17β-E2下调MG-63细胞IL-11mRNA及NF-κB蛋白质的表达。结论17β-E2下调MG-63细胞NF-κB、IL-6和IL-11的表达,此可能是妇女绝经后雌激素补充治疗的依据之一。 [Objective] To characterize the effects of 17β-estrodiol(17β-E2) on the expression of IL-6, IL-11 and NF-kB in human osteoblast line MG-63. [Methods] The expression of IL-6 was tested by RT-PCR, Northern blot and Western Blot, The expression of IL-11 was tested by RT-PCR and NF-kB was tested by Western Blot. [Result] 17β-E2 can down-regulate the expression of IL-6 mRNA and protein in MG-63 cells, 17β-E2 can downregulate the expression of IL-11 mRNA, protein and NF-kB protein in MG-63 cells. [Conclusion] The expression of NF-kB, IL-6 and IL-11 in MG-63 cells can be suppressed by 17β-E2, and this may support the estrogen replacement therapy in postmenopausal women.
