PNIPAM/HEMA固定化α-胰凝乳蛋白酶的研究

摘要 以聚N-异丙基丙烯酰胺/甲基丙烯酸-α-羟基乙酯为基质来固定α-胰凝乳蛋白酶,并对所制备的固定化酶的性能进行了多方面的研究,结果表明,酶促反应最适温度为30℃,最适pH=7.7,在30℃~45℃区间酶活力可以实现连续可调;经8次连续催化,固定化酶活力无明显下降;储存49天后,固定化酶活力变化不大,为进一步的研究固定化酶提供了必要的实验依据. In this paper,α - chymotrypsin immobilized in PNIPAM/HEMA copolymer gels was synthesized and its properties were studied. The measurements of enzyme activities were carried out in a series of experimentations. Results obtained showed that its optimum temperature was 30℃ and prefer pH was 7.7. lts enzyme activity could be adjusted continuously between a certain temperature (30℃ - 40℃ ), and was not reduced obviously after eight times continuous use for the catalyzing reaction. After forty - nine days for storage, the enzyme activity variation of the immobilized enzyme was not too big. All these will provide the necessary experimental basis for future research.
