摘要
探讨心力衰竭患者易患心房颤动(AF)的可能发生机制之一。采集左室功能减退组患者(n=18例)和左室功能正常组患者(n=18例)的右心耳组织,应用逆转录聚合酶链反应技术(RTPCR)以3磷酸甘油醛脱氧酶(GAPDH)为内参照,测定心房肌KV4.3α的mRNA表达水平,反应心房肌短暂外向钾电流(Ito1)的变化。结果:与左室功能正常组相比,左室功能减退组心房肌KV4.3α的mRNA表达减低46.20%(0.83±0.07vs0.45±0.09,P<0.01)。结论:左室功能减退患者心房肌KV4.3αmRNA表达降低。
To investigate the potential mecbanisms of atrial fibrillation in heart failure, right atrial appendages were obtained from 18 patients with decreased left ventricular function and 18 matched controls with normal left ventricular function. All patients were in sinus rhythm. The mRNA expression of I,o1 (KV4.3α) was measured by reverse transcripfion-polymerase chain reaction and normalized to the mRNA level of glyceraldehydes-3-phosphate dehydrogenase. Results: Compared to the control, the mRNA level of Itol (KV4.3α) in patients with decreased left ventricular function reduced by 46.20% (0.83 ±0.07 vs 0.45±0.09 ,P 〈0.01 ). Conclusion: The mRNA level of Itol (KV4.3α) is down-regulated in patients with decreased left ventricular function.
