葛根素对喹啉酸致海马神经元损伤的保护作用 被引量:1

摘要 目的研究葛根素对喹啉酸(qinolinic acid,QA)诱导原代培养乳鼠海马神经元兴奋毒损伤的保护作用。方法乳鼠海马神经元原代培养后用QA诱导损伤,采用MTT法测定细胞活力,同时测定培养上清液中乳酸脱氢酶 (LDH)活性和丙二醛(MDA)的含量。结果葛根素(3×10-4mol/L、10-4 mol/L)剂量能明显提高QA致损伤的原代培养乳鼠海马神经元的细胞活力,降低其上清液中LDH活性和MDA含量,与模型组比较差异均具有显著性(P <0.01)。结论葛根素(3×10-4moL/L,10-4 mol/L)对QA所致的原代培养乳鼠海马神经元损伤有明显保护作用。 Objective To study the protection effect of puerarin on the excitotoxicity induced by quinolinic acid(QA) on the cultured hippocampal neurons of new -borne rats. Metheds Excitotoxicity injury was induced by QA in cultured hippocampal neurons. The cell viability was measured by MTT assay, extracellular LDH and MDA contents was also measured. Results Puerarin with 3 ×10^-4·mol·L^-1 and 10^-4 mol·L^-1 could significantly reduce the death of the primary hippocampal neurons induced by QA, prevent leakage of LDH and decrease the contents of MDA. Conclusion Puerarin(3×10^-4mol·L^-1 and 10^-4 mol·L^-1) have protection effects on QA induced cytotoxicity in cultured hippocampal neurons of new-borne rats.
