DRB对人喉癌Hep-2细胞系中PK-CK2 mRNA表达的影响及其与细胞凋亡的关系

摘要 目的探讨5,6-二氯-1--βD-呋喃核糖苯并咪唑(DRB)对人喉癌Hep-2细胞系中蛋白激酶CK2(PK-CK2)mRNA表达的影响及其与细胞凋亡的关系。方法不同浓度的DRB作用于人喉癌Hep-2细胞后,采用半定量逆转录聚合酶链反应(RT-PCR)检测Hep-2细胞PK-CK2 mRNA表达水平的变化,采用流式细胞术检测Hep-2细胞凋亡率的变化。结果随着DRB浓度的增加或作用时间的延长,Hep-2细胞PK-CK2 mRNA表达水平呈下降趋势,实验组与对照组比较,差异有统计学意义(P<0.05),流式细胞术检测二倍体峰前出现凋亡峰,5、10、20、40、80μmmol/L DRB与Hep-2细胞共育2h后的细胞凋亡率分别为0.68%、1.17%、7.25%、10.40%、22.66%。结论DRB可下调Hep-2细胞PK-CK2 mRNA表达水平,诱导细胞凋亡,这些可能是DRB抗癌作用的重要机制之一。 Objective To detect the effect of 5, 6-Dichloro-1-β-D-ribofuranosyl-benzimidazole (DRB) on the expression of PK-CK2 mRNA and cell apoptosis in human laryngeal cancer Hep-2 cell line. Methods The cell apoptotic index and PK-CK2 mRNA expression levels in Hep-2 cells treated with different concentration of DRB were measured by Flow Cytometry (FCM) and RT-PCR at different time points. Results PK-CK2 mRNA levels in DRB-treated cells decreased as compared with those in untreated controls ( P 〈 0.05) with a concentration and time-dependent manner. Cell apoptotic indexes in Hep-2 cells treated with 5, 10, 20, 40, 80μmol/L DRB for 2 hours were 0.68 %, 1.17 %, 7.25 %, 10.40 % and 22.66 %, respectively. Conclusion DRB down-regulates PK-CK2 mRNA and induces apoptosis in Hep-2 cells, which may relate to its anticancer function.
