中药露蜂房蛋白对人红白血病细胞株K562细胞影响的实验研究 被引量:4

摘要 目的:研究中药露蜂房中蛋白成份抗白血病作用的机理,为中药露蜂房治疗白血病的临床应用和开发新的抗白血病药物提供实验和理论依据。方法:采用细胞培养和透射电子显微镜技术,观察中药露蜂房蛋白成份对人红白血病细胞(K562细胞)的生长抑制作用和形态结构的影响,采用免疫组织化学方法测定露蜂房蛋白对K562细胞凋亡相关信号转导蛋白NF-κBp65、-βcatenin及iNOS表达的影响。结果:(1)与生理盐水对照组比较,露蜂房蛋白处理组白血病细胞生长饱和密度降低,细胞增殖活性减弱(P<0.05);(2)露蜂房蛋白处理组白血病细胞呈典型的凋亡形态学改变;(3)与生理盐水对照组比较,露蜂房蛋白处理组白血病细胞中NF-κBp65、-βcatenin及iNOS表达减弱,NF-κBp65和-βcatenin细胞核内转导减少(P<0.05)。结论:中药露蜂房蛋白成份有明显抑制K562细胞的作用,其作用机制可能是通过调节凋亡相关信号传导因子NF-κBp65、-βcatenin及iNOS的表达,从而诱导白血病细胞凋亡。 Objective. To investigate the mechanism of the protein from Nidus Vespae on anti-leukemia effect and further provide experimental and theoretical evidence for clinical use of nidus vespae as the anti-leukemia medicine. Methods. The inhibitory effect and morphological changes of cultured K562 cells treated with water extracted protein from Nidus Vespae wre examined by transmission electronic microscope. The expressions of signal protein relating apoptosis including NF-κBp65、β-catenin and iNOS were detected in K562 cells via immunohistochemical method and image analysis system. Results.(1)Compared with saline control group, saturated density and ability of cell proliferation significantly decreased in experimental groups by curved line of proliferation(P〈0.05);(2) Under transmission electronic microscope, typical morphological changes of apoptosis were observed among leukemia cells in experimental groups; (3)Immunohistochemistry showed that in experimental groups, the expressions of NF-κBp65、β-catenin and iNOS obviously decreased in optical density(OD)analysed by the image analysis system. There was a significant difference between the experimental group and the saline control group(P〈0.05). Conclusion:The protein of Nidus Vespae can inhibit the proliferation of K562 cell significantly and regulate the apoptosis by inhibiting the expression of NF-κBp65、β-catenin and iNOS.
