食管癌组织中ToPoⅡ表达与癌细胞体外药敏反应关系的研究

Relationship between the expression of ToPoⅡ in esophageal carcinoma and their sensitivity to etoposide

目的探讨食管癌组织中ToPoⅡ表达与癌细胞体外药敏反应的关系。方法收集本院1999年至2003年手术切除的食管癌组织标本50例,经免疫组织细胞化学方法测定其组织中ToPoⅡ表达状态,用MTT法检测ToPoⅡ不同功能状态下各种肿瘤细胞对化疗药物Vp-16(足叶乙甙,ToPoⅡ抑制剂)的敏感性及差异。结果以Vp-16的IC50值计算,ToPoⅡ高表达组患者是低表达组患者的1/6,显示出较高的化疗敏感性。结论食管癌组织中ToPoⅡ功能状态可能对其化疗的敏感性有影响。

Objective To explore the relationship between the expression of ToPoⅡ in esophageal carcinoma and their sensitivity to etoposide. Methods The expression patterns of ToPoⅡ in 50 patients with esophageal carcinoma were examined by inmmunohistoehemistry. SP method using monoelonal antibody to their protein. Meanwhile,the sensitivity to etoposide in different ToPoⅡ expression levels was tested by MTT （ methyl thiazolyl tetrazolium） method. Results The sensitivity to etoposide was significantly different in different expression levels of ToPoⅡ in esophageal carcinorna, The IC50 of the Vp-16 in higher expression was significantly lower than that in lower expression. Conclusion The ToPoⅡ function status in esophageal careinoma directly influences their sensitivity to etoposide.
