摘要
目的探讨高压氧(HBO)暴露对离体小鼠腹腔巨噬细胞(PM)抗原提呈功能的影响。方法巨噬细胞分为:常压空气对照组(NA)、HBO组Ⅰ(HBO-Ⅰ)、HBO组Ⅱ(HBO-Ⅱ)以及HBO组Ⅲ(HBO-Ⅲ)。各组按相应方案暴露,相应时间点观察巨噬细胞吞噬功能、Fc受体功能、NO分泌量、TNF-α分泌量、淋巴细胞DNA合成状况以及Ia抗原表达的变化。结果经400 kPa 4 h HBO暴露,所测巨噬细胞的各指标均明显下降(P<0.05);经400 kPa 6 h HBO暴露,所测巨噬细胞的各指标亦明显下降(P<0.05和P<0.01),且较400 kPa 4 h HBO暴露变化更为明显。巨噬细胞经200 kPa 4 h HBO处理后,各指标变化不明显。结论一定压力和时程的HBO暴露能显著抑制小鼠腹腔巨噬细胞功能,体现了氧对免疫细胞的直接作用。HBO抑制巨噬细胞抗原提呈功能可能通过抑制其Ia抗原表达来实现。
Objective To study the effects of hyperbaric oxygen exposure on the antigen presentation function of mice peritoneal macrophages in vitro. Methods Macrophages were randomized in to 4 groups: normal air group (NA) , hyperbaric oxygen group Ⅰ ( HBO- Ⅰ ) , hyperbaric oxygen group Ⅱ ( HBO- Ⅱ ) , hyperbaric oxygen group Ⅲ ( HBO-Ⅲ ). Each group had its corresponding exposure schedule. And the phagocytosis activity,the Fc receptor function, the production of NO and TNF-α, the DNA synthesization of lymphocytes and the expression of Ⅰ region associated antigen were observed at the corresponding time. Results Compared with those in NA group,the phagocytosis activity, the Fc receptor function, the production of NO and TNF-α, the DNA synthesization of lymphocytes and the expression of Ⅰ region associated antigen were markedly inhibited in the HBO- Ⅱ group ( P 〈 0.05 ) and the HBO- Ⅲ group ( P 〈 0.05,P 〈 0.01 ) ;while they changed little in the HBO- Ⅰ group ( P 〉 0.05 ). Conclusions Hyperbaric oxygen exposure with a certain pressure-duration could remarkably inhibit various functions of mice peritoneal macrophages,which showed that oxygen could act on immune cells directly. And the inhibition of antigen presentation function of peritoneal macrophages after hyperbaric oxygen exposure might be related to the decrease of expression of Ⅰ region associated antigen.
