树突状细胞和小胶质细胞在脑胶质瘤自体免疫治疗中的作用机制 被引量:1

摘要 目的探讨树突状细胞(DC)和小胶质细胞(MG)在脑胶质瘤局部微环境中的关系及采用疫苗治疗后的变化。方法建立大鼠C6脑胶质瘤模型。采用C6细胞冻融抗原致敏DC制备疫苗,接种于造模第2天大鼠的皮下。以免疫组化法对大鼠脑内MG和DC的表达进行研究。结果治疗组DC表达均明显高于肿瘤组,而MG表达均明显低于肿瘤组。结论采用DC疫苗治疗可以增加局部微环境中的DC数量和改善免疫抑制状态,从而提高DC的功能。 Objective To investigate the relationship between dendritic cells (DCs) and microglias (MGs) in the microenvironment of brain glioma and their changes after treatment of DC vaccine. Methods Rat C6 brain glioma model was constructed. DCs were immunized with C6 cell freeze-thaw antigen to prepare vaccine being subcutaneously inoculated into rats on the second day of modeling. By using immunohistochemical method, the presentation of DCs and MGs was detected. Results The presentation of DCs in treatment group was significantly higher, while MGs significantly lower than that in tumor group. Conclusion DC vaccine can increase the number of DCs and improve the local immunosuppressive state.
