马桑内酯激活的星形胶质细胞条件培养液对正常大鼠脑内IL-1β的影响

摘要 目的揭示星形胶质细胞对大鼠脑内IL-1β的影响及其在癫痫发病中的作用。方法将马桑内酯(coriarialactone,CL)激活的星形胶质细胞条件培养液(astrocytic conditioned medium,ACM)注射入正常SD大鼠侧脑室,观察大鼠的行为变化,运用免疫组织化学及放射免疫分析的方法,观察大脑皮质、海马区域内白细胞介素1β(IL-1β)免疫反应的变化及脑组织匀浆、脑脊液内IL-1β含量的变化。结果ACM组大鼠在注射ACM后30min出现癫痫行为,2h恢复正常;免疫组织化学显示ACM作用后2h,IL-1β免疫反应阳性神经元数和平均吸光度值明显增加(均P<0.05),12h恢复正常水平;放射免疫分析显示ACM作用后2h大脑皮质、海马IL-1β含量均开始增加,4h达高峰(P<0.05),脑脊液中IL-1β含量则在ACM作用后2h即达到高峰。结论马桑内酯激活的星形胶质细胞条件培养液可增强大鼠脑内IL-1β的表达,并导致痫性发作。 Objective To explore the effect of coriaria lactone (CL)-activated astrocytic conditioned medium (ACM) on the intracerebral IL-1β and its role in epileptogenesis. Methods ACM was injected into lateral ventricle of SD rats and the behavioral changes were observed. Immunohistochemical SABC method was used to assess the changes of the expression of IL-1β in the cerebral cortex and hippocampus. The contents of IL-1β were determined by means of radioimmunoassay in homogenate of cerebral cortex and hippocampus and cerebrospinal fluid. Results Seizure was observed in ACM group 30 min after injection of ACM, but returned to normal 2 h later. The immunoreaction of IL-1β in hippocampus and cerebral cortex of rats 2 h after injection of ACM was stronger than that in the control group (P〈0.05), and the contents of IL-1β in homogenate of cerebral cortex and hippocampus and cerebrospinal fluid were higher than those in the control group (P〈0.05). Conclusion The ACM activated by CL can increase the expression of IL-1β and result in epileptogenesis.
