红花对局灶节段性肾小球硬化大鼠细胞外基质的影响 被引量:6

摘要 目的:观察红花对局灶节段性肾小球硬化(FSGS)大鼠细胞外基质(Extracellular matrix,ECM)的影响并探讨其部分作用机理。方法:30只健康雄性SD大鼠随机分为假手术组、模型组和红花组,采用一侧肾切除加两次尾静脉注射阿霉素的方法复制FSGS大鼠模型,8周后观察各组大鼠肾组织病理,并用免疫组织化学染色方法评价纤维连接蛋白(FN)、层黏连蛋白(LN)的表达。结果:模型组肾组织出现局灶节段性肾小球硬化,免疫组织化学提示FN、LN在肾小球及小管中表达明显(P<0·01);红花组FN、LN表达明显降低(P<0·01)。结论:红花能抑制FN和LN的表达,减少细胞外基质的合成及其在肾小球及小管间质中的过度沉积,延缓其病程进展。 Objective: to observe the effect of Honghua on extracellular matrix (ECM) in mice of foca^1 segmental glomerulosclerosis (FSGS), and to discuss parts of its functional mechanism..Methods: 30 health), male SD mice were randomly divided into sham operation group, model group and Honghua group, one side kidney removed and caudal vein injected with adriamycin twice so to make model mice of FSGS. 8 weeks later, pathology of nephridial tissues i.n each group were observed respectively, and the expressions of FN and LN were evaluated by means, of immunohistochemistry staining. Results: FSGS appeared in nephridial tissues of model group, and immunohistochemistry .indicated both FN and LN had obvious expression in renal glomerulus and tubule ( p 〈 0.01) ; the expressions of FN and LN in Honghua group decreased, significantly ( p 〈 0.01) Conclusion: Honghua can inhibit FIN and LN expressions, reduce ECM synthesis and its excessive deposit in renal glomerulus and tubule, and delay the course of disease.
