摘要
利用氯丙咪嗪能与 M 胆碱受体结合的性质,以氚标记二苯羟乙酸奎宁酯(~3H—QNB)为放射配基,建立了测定氯丙咪嗪血液浓度的放射配基结合法。研究表明,本方法最小可检测12ng/ml,回收率为103.4%;批内误差8.5%,批间误差15.6%;反应误差系数为0.042。放射配基结合法具有较稳定、灵敏度高、简单方便、快速又经济的特点,其最大优点是能够测定血液中代表总药理活性的药物及其代谢产物的含量。
Objective:To study the use of radio—ligand assay in determining serum clomipramine level. Method:The assay was based on the principle that,using 3H—QNB marked by tritium as the radio- ligand,serum clomipramine competed for the binding of M cholinergic receptors in rat brain.Results: The radio-ligand assay detected serum clomipramine level at concentrations as low as 12ng/ml.The accuracy was 103.4%.The intra-assay and inter-assay coefficients of variation were 8.5% and 15.6% respectively.The response error relationship was 0.042.Discussion:The radio—ligand assay was sensitive,fast and yet simple and economical.It can profitably be used to detect serum levels of oth- er drugs as well as their metabolites.
