内毒素急性肾损伤时肾脏ELAM-1mRNA的表达及其意义

摘要 利用地高辛标记CRNA探针原位杂交技术检测内毒素性急性肾损伤大鼠肾组织内皮细胞白细胞粘附因子-1(ELAM-1)mRNA的表达.结果发现;受检肾组织毛细血管网及小静脉血管内皮细胞呈现ELAM-1mRNA的广泛表达,其时程变化为给予内毒素后1h已出现弱表达,3~6h达高峰,24h内衰城近无.这种表达提示了内毒素性急性肾损伤早期肾小球毛细血管内大量中性粒细胞(PMN)滞留聚集的重要分子基础,对于解释中毒性休克时急性肾功能衰竭的发生机理有重要价值. Background There is ample evidence for a pathogenic role of neutrophils in acute kid-neys injury induced by endotoxins. Neu-trophil sequestration in the renal microvas-culature is considered a critical early event.The mechanisms by which the endotoxins cause neutropenia and neutrophil sequestra-tion in the kidneys are poorly understood.Neutrophil adherence to pndothelium is par-tially mediated by the expression of en-dothelial leukocyte adhesion molecule-1 (E-LAM-1) on endothelial cells activated by a-gents such as endotoxins. The purpose of this experimental study was to show the ex-pression and distribution of ELAM-1 of re-nal tissue in endotoxemia.Experimental design ELAM-1 mRNA ex-pression in renal tissue of endotoxin-induced acute injury was examined by the use of digoxigein labelled RNA probe in situ hy-bridization techniqe.Results The results showed that there was widespread expression of ELAM-1 mRNA in vascular endothelial cell of glomerular and small veins. The expression was weak 1 hour after intravanous administration of en-dotoxin, and was maximal during 3 to 6 hours and decreased within 24 hours.ConcIusions The results suggest the im-portant molecular basis of tremendous PMN sequestration in glomerular in the early pe-riod of acute kidneys injury,and give a bet-ter elucidation on the mechanisms of acute renal failure which occur in septic shock.
