马立克氏病病毒pp38基因产物对其上游双向启动子活性的增强作用 被引量:2

摘要 为了研究鸡马立克氏病病毒(MDV)pp38基因和1.8 kb mRNA转录子基因间一双向启动子的活性,本研究以增强型绿色荧光蛋白(EGFP)为报告基因,构建了启动子以pp38方向转录的重组质粒pP(pp38)-EGFP和以1.8 kb方向转录的重组质粒pP(1.8-kb)-EGFP.将这二个重组质粒分别转染鸡胚成纤维细胞(CEF)、MDV rMd5克隆株感染的CEF(rMd5-CEF)以及pp38基因缺失的MDV rMd5Δpp38克隆株感染的CEF(rMd5Δpp38-CEF),结果只有在转染rMD5-CEF样品中才能检测到EGFP的表达;将上述二个EGFP重组质粒分别与能表达pp38的重组质粒pcDNA-pp38共转染CEF和rMd5Δpp38-CEF,在CEF中仍检测不到EGFP的表达,而在rMd5Δpp38-CEF形成的空斑周围,能检测到EGFP的表达.结果提示pp38是该双向启动子发挥作用的必要条件,但该启动子活性的发挥仍需除pp38以外的其他因子的共同作用.将pcDNA-pp38质粒和pp24表达质粒pcDNA-pp24,以及EGFP表达质粒共转染无MDV感染的CEF时,能检测到EGFP的表达.核酸阻滞试验表明,pp38必须和pp24共表达时,才能和启动子结合,表现出阻滞现象.说明pp38和pp24可能以聚合体形式与启动子结合并增强启动子的转录活性.还证明了该双向启动子在1.8 kb mRNA转录子方向的启动活性显著高于pp38基因方向.
