诱导的与凝集素受体相似的c-rel基因的分子克隆和鉴定

摘要 目的进一步了解c-rel对免疫系统的调控功能特征。方法采用再现差异分析法(representative difference analysis,RDA)分析从野生型和c-rel敲除老鼠所获mRNA。结果发现一个新基因,其表达依赖于完整c-rel,命名为淋巴细胞衍生的C-型凝集素(LCL)-1。LCL-1属Ⅱ型跨膜蛋白,膜外区含一个糖基识别域。该糖基识别域与其他C-型凝集素受体(包括CD69)所携带的糖基识别域具有高度同源性。LCL-1基因位于小鼠6号染色体的NK细胞基因复合体内,编码至少4种可选择的拼接亚型。有趣的是,LCL-1除表达于B细胞,也在不成熟和成熟树突状细胞表达,尤以后者表达水平更高。结论发现一个NKC家族新成员,首次证明NKC家族的凝集素样受体是c-rel的下游基因。 Objective To further characterize c-tel regulatory function in the immune system. Methods We performed a representational difference analysis(RDA) on mRNAs derived from B-lymphocytes of wild type (WT) and c-rel knockout(KO) mice. Results We identified a novel gene designated lymphocyte-derived C-type lectin ( LCL)- 1, its expression is dependent on intact c- rel. LCL-1 is a type Ⅱ transmembrane protein with a single extracellular carbohydrate recognition domain(CRD) that exhibits homology to C-type lectin-like receptors including CD69. LCL-1 gene is localized at the NK germ complex(NKC) on mouse ehromosome 6 and encodes at least four alternatively spliced isoforms. In addition to its expression in B cells, LCL-1 is also expressed in immature as well as mature dendritic cells(DC), with higher expression level seen in mature DC. Conclusion Our findings offer more information to NKC family and identify a lectin-like receptor of the NKC family as a c-rel target gene.
