脊髓半切损伤后髓鞘碱性蛋白及胶质纤维酸性蛋白的表达及意义 被引量:6

摘要 目的研究大鼠脊髓半切损伤后后肢运动功能恢复情况及损伤远端星形胶质细胞和近端少突胶质细胞反应性增生关系,探讨损伤后星形胶质细胞和少突胶质细胞反应性增生在脊髓损伤修复中的作用.方法将35只SD大鼠随机分成7组:对照组5只;实验组6组,每组5只,分别是伤后1天组,3天组,7天组,14天组,21天组,28天组.实验组动物均行T9左侧半切,分别于伤后1,3,7,14,21,28 d行后肢运动功能联合评分(CBS)及Basso-Beattle-Bresnahan(BBB)评分.每次评分后处死5只大鼠,进行HE染色及GFAP、MBP免疫组织化学检测.结果伤后后肢均有不同程度的恢复,1~2周时恢复幅度最大,2~3周时后肢运动功能继续恢复,3周时BBB评分最高达12分,3~4周运动功能无显著性恢复,损伤后1 d损伤远端3~6 mm处GFAP阳性星形胶质细胞开始增生肥大,3~4 d灰质中星形胶质细胞明显增多,2周时达到高峰,损伤近端3~6 mm处少突胶质细胞的增生反应过程与星形胶质细胞相似.结论脊髓损伤后近端少突胶质细胞和远端星形胶质细胞反应性增生可能有助于损伤的修复和再生. Objective To observe the relation oligodendrocyte reaction and astrocyte reaction of of the recovery process of hind limb function and the caudal to the injury site after spinal cord hemisection. investigate the effect of oligodendrocyte and astrocyte reactive hyperplasy after spinal cord injury. Methods 35S-D Rats were randomly divided into 7group, experiment group(n = 30) and the control group( n = 5), the experiment rats were hemisected at T9 plane and a 3 mm spinal cord tisse was removed for forming a 3mm gap. The functional recovery of the hind limb was evaluated using CBS(combined behavioral score) and Basso-Beattle-Bresnahan (BBB)1, 3, 7, 14, 21, 28 days after injury. At every time, site after hind limb function scored, 5 rats were sacrificed, experiments were done on histology, immunohistochemistry, and counting the labeled cells in the gray matter. Results The hind limb function of the injured rats recovered at different degrees, the most extent recovery occurred during the time site 1- 2 week, recovery continued from 2 to 3 week and, BBB was up to 12 at the end of the third week, but there was no significance recovery during 3-4 week. the astrocyte caudal to the injury plane began hyperplasy and hypertrophy;astrocyte in which GFAP in expression was positive the gray matter increased obviously from 3 days to 14 days after hemisection. The expression of MBP is same as that of GFAP. Conclusion After active oligodendrocyte and astrogliosis accompanion important role in self-repair and regeneration. spinal cord injury there is obviously increase in rewith hind limb motive recovery which may play an
