摘要 目的:通过观测缺氧后小鼠肝细胞的形态结构及肝糖元量的多寡,探讨不同缺氧状态下小鼠肝组织结构的变化。方法:用间歇性重复缺氧模型和持续性缺氧模型小鼠,取肝组织作HE染色和PAS法组化染色,观察肝组织结构变化;对肝糖元作定量分析。结果:(1)缺氧组小鼠肝细胞出现混浊肿胀,部分肝细胞核变小;(2)PAS反应显示,持续缺氧小鼠肝细胞糖原颗粒明显减少。结论:缺氧时氧自由基明显增加,肝细胞供能障碍,出现结构损伤,但间歇性重复缺氧可能通过某种途径启动内源性的保护机制,从而减轻缺氧造成的损害。 Object: To research the changes of mouse hepatic tissue structure under different hypoxia conditions by observing the mouse hepatocyte structure and glycogen. Methods: We obtained the hepatic tissue fromn the intermittent hypoxia mouse modal and continual hypoxia mouse modal. The hepatic tissue was stained by hematoxylin eosin (HE) and periodic acid Schiff (PAS). Observe the changes of mouse hepatic tissue structure and glycogen. Results: (1)The mouse hepatocyte appeared albuminous swelling and some nucleus of hepatocyte became smaller in hypoxia group. (2)The results of PAS reaction showed that the glycogen of hepatocyte decreased obviously in the continual hypoxia mouse model. Conclusion: Hypoxia results in a serial changes, oxygen free radical increased evidently, the obstacle of bepatocyte energizing and the damage of hepatic tissue structure. The less damage in intermittent hypoxia group might be caused through a certain endogenous protective mechanism.
