盐酸伊曲康唑胶囊的人体生物等效性研究 被引量:8

摘要 目的:评价单剂量口服2种伊曲康唑胶囊的人体药动学和生物等效性。方法:采用随机、交叉试验设计。20名男性健康志愿者单剂量口服200mg伊曲康唑,用高效液相色谱一荧光检测法测定其体内血药浓度。结果:受试制剂和参比制剂t1/2分别为(29.3±5.62)、(29.3±5.81)h,Cmax(81.4±60.0)、(77.8±45.2)μg/L,tmax(3.9±0.70)、(4.2±0.70)h,AUC0~72(1199.4±649.6)、(1174.3±701.9)(μg·h)/L,AUC0~∞(1414.0±815.2)、(1386.1±735.8)(μg·h)/L,经方差分析和双单侧t检验,主要药动学参数无差异,但tmax存在差异;受试制剂的相对生物利用度为(105.3±23.4)%。结论:2种伊曲康唑胶囊具有生物等效性。 OBJECTIVE: To evaluate the human body pharmacokinetics and bioequiavailability of two kinds of oral single dose of hydrochloric itraconazole capsules. METHODS: A randomized, crossover study of 20 healthy volunteers receiving single oral dose of 200 mg itraconazole was conducted with in vivo blood concentrations determined by HPLC- fluorescence detection. RESULTS: The pharmacokinetic parameters for the testing itraconazole and the reference itraconazole were as follows, t1/2 were(29.3± 5.62)h and(29.3± 5.81)h, respectively; Cmax were(81.4± 60.0)μg/L and(77.8± 45.2)μg/L, respec- tively; tmax were(3.9± 0.70) h and(4.2± 0.70) h, respectively; AUC0-72 were(1 199.4± 649.6) (μg · h)/L and (1 174.3± 701.9 ) (μg · h)/L, respectively; AUCo-= were(1 414.0± 815.2) (μg · h)/L and(1 386.1 ± 735.8) (μg · h)/L, respectively. there were no significant differences in main pharmacokinetics parameters between 2 preparations, except in tmax from analysis of variance and one - side & two - sides t - tests. The relative bioavailability of trial itraconazole capsule was ( 105.3 ± 23.4) %. CONCLUSION: These two kinds of itraconazole capsules are bioequivalent.
