摘要
目的 动态观察脑脊液对骨髓基质细胞向神经元方向分化的作用。方法 采用Percoll液分离大鼠骨髓基质细胞,体外扩增。当第2或第3代细胞生长达60%~70%融合时,加入不同病人脑脊液,分别于不同时间观察其形态学改变,取诱导后的细胞作免疫细胞化学鉴定。结果 在颅咽管病人脑脊液中骨髓基质细胞的分化速度较快,且发生政变的程度较大;而在脑外伤积水的病人脑脊液中骨髓基质细胞的分化速度相对较慢,且发生改变的细胞数较前者少。免疫细胞化学染色NSE呈弱阳性,NF呈弱阳性。结论 脑脊液可诱导骨髓基质细胞向神经元方向分化。
Objective To observe the effect of cerebrospinal fluid(CSF) on bone marrow stromal cells(BMSCs) differentiated into neurons. Methods BMSCs were isolated from rat bone marrow with Percoll. When the cells(from passage 2 or passage 3) confluent about 60%- 70%, the media were replaced with CSF. The morphology of the subpopulation cells of BMSCs was observed at differ ent time. Cells were fixed for immunocytochemistry. Results Defferentiation of BMSCs that had mor phologieally changed was quilker in craniopharyngioma patient's CSF than that in traumatic brain injury patient's CSF. The differentiated cells expressed less possitively NSE and NF, the markers of neuron. Conclusion CSF could induce BMSCs to differentiate into neurons.
