摘要
在高效毛细管电泳中,pH值是生物产品分离中的重要参数,它不仅影响蛋白质的迁移速度,而且影响电渗速度,进而影响高效毛细管电泳的分高效率。研究了高压电场下毛细管中介质pH值对电渗速度的影响,并建立了氢离子表面吸附数学模型。用实验数据拟合了模型参数。该模型与实验结果较好地吻合,并能阐明介质pH值与电渗速度的线性关系。
An important parameter for bioproduct separation in the high performance capillary electrophoresis is pH value, which effects not only on the migratory velocity, but also on the electroendoosmosis and further on the separation efficiency. The effect of pH value of medium on the electroendoosmosis flows in the electric field applied high voltages was studied in detail and a surface adsorption model of hydrogen ion was developed. The parameters of the model were estimated by experimental data. It was showed that the model fitted the experimental results very well and can explain the linear relationship of pH values to the electroendoosmosis flows.
