摘要
采用大鼠和犬,经连续3mon口服给药,对雷公藤微囊(TWH)的毒性进行了研究。5mg·kg-1TWH对大鼠和犬均不表现任何毒性作用,TWH15mg·kg-1对大鼠,10mg·kg-1对犬有一定的毒性作用,但停药后,可以恢复;高剂量下(大鼠35mg·kg-1,犬15mg·kg-1),大鼠血糖、血尿素氮及犬的碱性磷酸酶活性显著增高,犬血总蛋白及血白蛋白含量显著下降,病理组织学检查也表明大鼠及犬的心、肝、肾等多种脏器有明显损害,但上述毒性停药后基本可以恢复,也未发现有明显的延迟毒性,表明高剂量TWH所产生的毒性是可逆的。
The long-term toxicities of tripterygium wilfordii hook(TWH) were studied by oral administration on 90 consecutive days to rats and dogs respectively.TWH 5 mg·kg-1 to dogs and rats did not show any toxicity. 10 mg·kg-1 to dogs and 15 mg·kg-1 to rats had certain toxic action, but the animals could recover from these toxicities after being ceased administration TWH for 14 days.In high doses of 15 mg·kg-1 to dogs and 35 mg·kg-1 to rats, TWH could significantly increase the content of blood urea nitrogen and sugar in rats and the activity of plasma alkaline phosphatase in dogs,meanwhile,TWH reduced the content of plasma total protein and albumin in dogs. Histological examination showed obvious toxicities in heart,liver,kidney and other important organs of both rats and dogs,the animals could also get over.The results indicated that the toxicities induced by high doses of TWH were reversible.
