摘要
目的:研究红花黄色素在大鼠各肠段的吸收动力学特征。方法:采用大鼠在体肠段灌流实验,主要从药物浓度、pH值、吸收部位等3方面对红花黄色素的肠段吸收特性进行研究。结果:在5μg·mL^(-1)~20μg·mL^(-1)浓度范围内药物浓度对吸收速率常数无影响;在pH7.8~pH5.4内药物吸收不受pH值影响;各肠段的吸收速率常数按空肠、十二指肠、回肠、结肠顺序依次下降,分别为0.1116,0.0753,0.0739,0.0185h^(-1)。结论:药物吸收符合一级动力学特征,吸收机制为被动扩散。
Objective: To investigate the absorption characteristics and mechanism of safflow yellow for various intestinal segment. Method: The intestine in rats was cannulatod for in situ recirculation. The effects of drug concentration, pH value and absorption site on safflow yellow absorption were studiod. Results: The concentrations of safflow yellow from 5μg·mL^-1 to 20μg·mL^-1 and the pHof drug solution from pH 7.8 to pH 5.4 had no distinctive effect on the absorption kinetics. The absorption rate constant (Ka) were 0.1116, 0.0753, 0.0739, 0.0185h^-1, at jejtmum, duodenum, ileum and colon, respectively. Conclusion: The absorption of safflow yellow was a first-ordar process with the passive diffusion mechanism.
