摘要
目的:探讨高速枪弹伤时产生的强压力波对血管内皮细胞内皮素(ET-1)的释放及基因表达的影响。方法:以高速枪弹致伤犬腹部以及高速枪弹压力波致伤培养人脐静脉内皮细胞为实验模型,采用放射免疫及斑点杂交的方法,从整体、细胞、基因不同层次观察内皮细胞ET-1释放及表达的特点。结果:致伤犬和培养的内皮细胞在压力波作用下,ET-1释放和基因表达的变化具有相似的趋势,出现即刻和延迟反应,即伤后10分钟至6小时ET-1释放明显增加,mRNA(ET-1)转录增强则出现在伤后4~6小时。结论:压力波是导致ET-1释放及基因表达紊乱的重要力学因素,ET-1mRNA转录增强可能是压力波对ET-1调节的延迟效应的分子病理学基础。
Aim: To investigate the changes in an release and gene expression of Endothelin (ET 1) from endothelium affected by a stronger pressure wave after high velocity bullet wounds. Methods: It was taken for the experimental models that the canine abdomens were shot with the high velocity bullets and the endothelial cells of the cultured human umbilical veins were struck immediately by a pressure wave after high velocity bullets impacting water. The release and gene expression of ET 1 in the endotheluim of the wounded dogs and the cultured cells were determined with radioimmunoassay (RIA) and dot plot hybridization. Results: The immediate or delayed effects were seen in the affected endothelial cells both in vivo and in vitro after wounding. The release of ET 1 from the cells increased from 10 minutes to 6 hours, and an upregulation of gene expression from 4 to 6 hours. Conclusion: The pressure wave induced by the high velocity bullets may be an important factor for the disturbance of release and gene expression of ET 1. The upregulation of ET 1 mRNA may be a molecular pathogenic factor for the delayed effect on the regulation of ET 1.
