摘要
目的:探讨慢性乙型肝炎者树突状细胞(DCs)的功能状态及机制。方法:以直接荧光法进行表型分析、CCK8法测定DCs对同种异体淋巴细胞的刺激作用、AnnexinV/FITC法测定细胞凋亡率进行分析。结果:慢性乙型肝炎患者外周血单核细胞经诱导分化形成的DCs,与正常人相比,表达协同刺激分子CD80、CD86水平均较低;刺激同种异体淋巴细胞增殖能力也低于正常人;DCs凋亡率明显增高。结论:慢性乙型肝炎患者外周血的DCs免疫功能处于抑制状态,而HBV感染导致的DCs凋亡增多可能是引起DCs功能障碍的原因之一。
Objective: To study the functional status of the dendritic ceils (DCs) derived from PBMC in the patients with chronic hepatitis B and its probably mechanism,a comparison was made in the differences in phenotypes and the stimulating capacities of allogeneic lymphocytes and the apoptosis of the DCs between patients with chronic hepatitis B and those of the normal controls. Methods: The DCs were phenotyped by cytometric analysis,the functional status of these ceils was estimated by the stimulating reactions of allogeneic lymphocytes and the apoptosis of these ceils was estimated by AnnexinV/FITC. Results: The expressions of CD80, CD86 molecules on the DCs from patients with chronic hepatitis B were lower in comparison with those of the normal controls; the capacity of DCs to stimulate the allogeneic lymphocytes was weakened in patients but the apoptosis rate of the DCs of patients was significantly greater. Conclusion:The functional status of the DCs of patients with chronic hepatitis B is inhibitory and the more distinguished apoptosis of the DCs from patients with chronic hepatitis B may be the one of the probable mechanisms of its dysfunction.
