摘要 目的:观察刺五加皂苷(ASS)对体外培养的缺血运动神经元生长有无保护作用;并初步探讨其保护作用机制。方法:对胚鼠运动神经元进行原代分离培养,建立缺血缺氧诱导的神经元细胞凋亡模型,用MTT法测定神经元细胞活性;检测LDH释放量来观察ASS对神经元细胞膜稳定性的影响。结果:刺五加皂苷能提高缺血缺氧神经元的细胞活性,降低缺血缺氧神经元LDH的释放量,稳定细胞膜,对缺血缺氧神经元有保护作用。结论:ASS对体外培养缺血缺氧损伤的运动神经元有明显的保护作用,可能与增强细胞膜的稳定性有关。 Objective :To determine whether Acanthopanax Senticosus Saponins (ASS) has protective effects on the anoxia of the spinal cord and to explore the molecular mechanism of the protective effects. Methods:The SMNs was dissociated from embryonic 15 day rats and cultured in vitro.The model of apoptosis induced by anoxia was established on cultured motor neurons.Neuron viability was measured by MTT assay and LDH (lactate dehydrogenase) release.The changes of morphology were observed by phase-contrast microscopy Results:ASS could enhance the neuronal viability and decrease the LDH release. Under phase-contrast optics, many hypoxic neurons were found shrunken with no processes and the sister cells pretreated by ASS were found less similar. Conclusions: ASS has affirmnative protective effects on hypoxic neurons, which is related to the stabilization of cell membranes.
