Viral hepatitis is one of the serious public health problems all over the world. Prevention and treatment of hepatitis B is a vitally important subject. The epitope of Pre-S protein onthe out membrance of HBV is essential for binding of HBV to its target cell surface. IFN is oneof the cytokines with multiple biological functions in human body and can play an important rolein prevention and treatment of HBV. With PCR technique, we amplified the whole genes of IFNα A and HBV Pre-S. In order to manupulate the cloning and expresstion of a fusion gene, corresponding endonuclease sites, protective sites, initiation and termination codon were designed atthe 5'terminal of the PCR primer. A clone of fusion gene IFN α A-HBV Pre-S was constructedand sequenced, which could be used for expressing a fusion protein with both biological activitiesof IFN and immunogen icity of HBV Pre-S.
