摘要
目的:利用RT-全套式PCR对胎儿各器官和组织中VEGF转录丰度进行研究。方法:检索人VEGFmRNA的序列,设计、合成上下游引物,提取总RNA,使用RT的方法合成cDNA,通过全套式PCR对VEGF特异性条带(3 7 4 bp)进行鉴定。结果:经过agarose电泳检测,人胎儿期各器官和组织cDNA的PCR扩增产物中,均出现一条不同灰度:3 7 4 bp的特异性条带。结论:VEGF在胎儿各器官和组织中转录丰度顺序为:大脑、心脏、肾脏、骨骼肌、肝脏、肺脏。
Objective: To demonstrate the VEGF transcriptional discriminations conceiving in various organs in fetus by using the regular nested-PCR. Methods: The forward and backward primers were designed optimally and respectively according to the published human vascular endothelial cell growth factor sequences from GENEBANK. The total RNA were extracted. The human fetus HIF cDNA was obtained by the RT - PCR, The unique band-374bp appeared on the gel was identified by the nested - PCR. Results: Calibrating the gel with the Ultraviolet Transilluminator, the wanted 374bp band equally appeared in different lighting density. Conclusion: The final order of human fetus VEGF gene transcription - level is brain, heart, kidney, Skeleton - muscle, liver, lung.
