摘要
目的:建立一种用蛋白质芯片检测血清中弓形虫、巨细胞病毒、风疹病毒、单纯疱疹病毒(TORCH)抗体的新方法。方法:采用玻片表面修饰琼脂糖膜制备蛋白质芯片,以荧光标记抗体,检测血清中TORCH IgM抗体。结果:所制备的蛋白质芯片可检测到血清中微量TORCH抗体的存在,荧光强度和抗原质量浓度呈线性关系,抗体之间交叉反应可忽略不计。该芯片法与ELISA试剂盒检测阴阳性符合率均100%。结论:蛋白质芯片技术检测孕妇TORCH感染,具有高通量、微型化、操作简便等优点,可望应用于临床中。
Objective To develop a new method of TORCH protein microarray for detecting TORCH infections. Methods The antigen were printed on glass slide modified with the agarose films. The serum antibodies against the TORCH antigen were detected by using fluorescence labeled secondary antibodies. Results TORCH protein microarray could detect minimum antibodies against the TORCH antigen. The cross-reactivities of the anti-IgM goat were negligible. 100% concordance were observed between microarray and ELISAs. Conclusion The TORCH protein microarray,having the advantages of high throughput, convenience, micromation, will be used in clinic in the future.
