一株菲降解细菌的培养条件优化及特性研究 被引量:1

摘要 通过选择性富集培养.从沈抚灌区石油污染土壤中分离到1株菲降解细菌L2,该菌株能以菲为唯一碳源和能源生长。通过对L2菌株培苯基厦培养条件优化,确定其最佳培齐基为玉米粉4%,蛋白胨1%,NaC11%,酵母膏0.5%;最佳培养条件为pH值7.0~7.2,温度28-30℃,150mL三角瓶装液量50mL。在此基础上测定了L2菌株在不同浓度菲中邻苯二酚2,3双加氧酶活力。结果表明,随着菲浓度从50mg·mL^-1增加到350mg·mL^-1,邻笨二酚2,3-双加氧酶活力呈先增加后减小趋势。 A phenanthrene-degradating strain L2 was isolated from oil-contaminated soil in Shenfu irrigation area by selective enrichment culture. The strain was able to utilize phenanthrene as the sole carbon source for growth. It was suggested by the optimization of cultural substrates and conditions that the optimum cultural substrates were: maize starch 4%,peptone 1%,yeasts exact 0.5%,NaCl 1%,and the optimum cultural conditions were : pH 7.0-7.2,28-30℃, 50 mL inventory/150mL flask. Meanwhile, the catechol 2,3-dioxygenase activity under different phenanthrene concentrations were determined. It was shown that with the increasing of phenanthrene concentrations from 50 mg · mL^-1 to 350 mg·mL^-1 ,the activity of catechol 2,3-dioxygenase in creased at first and decreased afterwards.
