摘要
目的探讨提纯人胆肝细胞再生因子(HHGF)方法及对小鼠中毒性肝炎的治疗作用。方法人胚肝细胞裂解物加热后分别经稳膜;超滤,再经DEAE离子交换层析和FRLCmonoQ阴离子交换色谱,最后经SDS-PAGE分析.用3H-TdR渗入法在人胚肝细胞和SMMC-7721人肝癌细胞株中检测HHGF活性.尾静脉注射HHGF治疗小鼠CCl4,中毒性肝炎。结果纯化的HHGF较纯化前相对活性提高2万倍.HHGF可使小鼠CCl4中毒性肝炎升高的ALT和SBIL在96h时接近正常,死亡率从对照组60%降至为0。并有促进肝细胞DNA合成作用.组织学检查HHGF有促进肝细胞再生和治疗肝损伤作用.结论HHGF对促进肝细胞再生。
Objective To research purification the human hepatic growth factor (HHGF) and the therapeutic effect for the toxic hepatitis of mice.Methods After being heated, HHGF was highly purified from the cleavage of the hepatic cells of human embryon, by ultrafiltration with filter membrance and by treated with the ion exchange chromatography of DEAE and kyoto Q ion exchange chromatography of FR LC.The H UGF activites were measured in hepatic cells and the SMMC-7721 human hepatic cancer cell-line with the AH-TdR incorporation. By injection of HHGF into the tail vein,the toxic hepatitis of mice was treated. Results The relative activity of the purified HHGF raised 20000 times more than before purification. Treating the CC14toxic hepatitis of mice with HHGF, the raised ALT and SAIL approached normal levels after 95 h,and the death rate decreased from 60% to 0. HHGF can increase the reproduction of liver cells,and promote the synthesis of the DNA of liver cells. It appeared that HHGF has an treatment effect on liver injury.Conclusion HHGF has an obvious effect on increasing the reproduction of the liver cells, releasing the injury of liver in the toxic hepatitis.
