摘要 细胞内多胺合成增加在维持细胞恶性表型中起重要作用,抑制其合成可负调控癌细胞的增殖。本文用多胺合成限速酶———鸟氨酸脱羧酶的抑制剂α二氟甲基鸟氨酸(DFMO)限制多胺合成,观察其对HL60细胞生长的影响。发现DFMO使HL60细胞向粒细胞分化并进行凋亡。提示,抑制多胺合成在细胞分化和凋亡中有重要作用。 The increasing bio synthesis of polyamines plays an important role in the development of malignancy of tumor cells.The depletion of intracellular polyamines by inhibiting the bio synthesis of polyamines can cause tumor cells growth arrest and reverse tumorigenicity.Using DFMO,an inhibitor of ODC enzyme which catalyzes the bio synthesis reaction of polyamines,the purpose of the study was to investigate the characteristic changes in the growth of HL 60 cells with depletion of intracelullar polyamines.The results showed that polyamines depletion induced HL 60 cells to differentiate into granulocytes and subseqently undergoing apoptosis.These data indicated that inhibition of the bio synthesis of intracelullar polyamines produced important effect on HL 60 cells proliferation,differentiation and apoptosis.Hence,block of the polyamines bio synthesis metabolism might be an approach to treat cancer.
