摘要
结扎小鼠一侧颈总动脉,建立急性脑缺血动物模型。术前腹腔给予人胚胎脑组织提取的生物活性多肽类物质神经肽,术后一小时测定脑组织中乳酸脱氢酶(LDH)活性,同时进行组织学和超微结构观察。结果显示给药组小鼠脑组织LDH活性明显高于未给药组,给药组小鼠脑组织脱水不明显,神经细胞中无明显空泡,细胞内超微结构改变较未给药组轻。本研究说明种经肽对脑组织急性缺血具有保护作用。
Neuropeptide(NP)is a biological product extracted from human fetus brain tissue. Several nerve growth factors were thought to the be included in this mixture. It was rePOrted that NP could promot the propagation of central and outer neurons. The effect of NP on acute cerebral ischemia is still,unknown.We have successfully developed the acute brain ischemia model in mice by ligation of unilateral arteria carotis communis. To determine the effect of NP, the mice were given NP by intraperitoneal injection 30 minutes before ligation.The brain tissues of injected mice were harvested one hour after ligation. Fetocerebral injection(FI)an analogue which is being used in clinical therapy,was employed in this experiment as a positive control. Lactate dehydrogenase(LDH)activity,histological morphology, and ultrastructure of the injected mice brain tissue;had been investigated.Our experiment showed LDH activities decreased sharply in non-injected mice brain,histological morphologies and ultrastructures were also demaged fiercely,however,those changes were obviously improved in the injected mice. There was a significant difference between the LDH activities of non-injected mice and of injected micecompared by t test (p<0. 01), but no such a significant difference could hi found between NP group and FI group (p>0.0.5).According to the results,we believe that NP can protect the central nerve sytem from acute cerebral ischemia,and it should be a helpful pharmaceutical for the treatment of brain ischemic diseases.
