大鼠动脉内少量冷盐水输注的神经保护作用研究

摘要 目的研究大鼠动脉内少量冷盐水输注的神经保护作用。方法应用内插有尼龙线的PE-50导管制作大鼠大脑中动脉(MCA)缺血3 h模型,在再灌注前5 min向MCA缺血区输注3.5 ml 10℃盐水,然后实施再灌注。对照组仅仅制作缺血3 h/再灌注48 h模型,未输注冷盐水。再灌注48 h后,用TTC染色法分别测定两组的脑梗死体积。在缺血后2 h和再灌注后48 h进行神经功能缺失评分。结果冷盐水输注组能显著减少脑梗死体积(23.8±3.0%),与未输注组相比(44.3±1.4%)差异有显著性(P<0.01)。再灌注前后两组的神经功能缺失评分差异有显著性(P<0.01)。结论动脉内少量冷盐水输注可显著减少脑缺血/再灌注后的脑梗死体积,产生神经保护作用。 Objective To study the neuroprotective effect on rats with cold saline infusion intra- arterially. Methods A PE-50 catheter with a silicon was used to block the middle cerebral artery (MCA) of rats for 3 hours in cold saline infusion group, and locally infuse the MCA - supplied territory with 3.5 ml 10℃ saline for 5 minutes before reperfusion. Control group was just M CAO/reperfusion models without cold saline infusion. Infarct volume was measured after 48 hours of reperfusion. Neurological deficit scoring was measured 2 hours after cerebral ischemia and 48 hours after reperfusion in both two groups.Results Cold saline infusion group significantly reduced cerebral infarct volume (23.8 ± 3.0% ), and there was a significant difference (P 〈 0.01 ) compared with none infusion group (44.3 ± 1.4% ) after 48 hours of reperfusion. There was a significant difference ( P 〈 0.01 ) between two groups in neurological deficit scoring. Conclusion Intra - arterially cold saline infusion can significantly reduce infarct volume and induce neuroprotection in cerebral ischemic/reperfuscd rats.
