摘要
目的了解中国HIV/AIDS患者tat第一外显子的基因序列的特征和变异特点,探讨其与HIV-1感染者疾病是展之间的关系。方法选取40例辽宁和吉林HIV感染后临床进程不同的人抽取外周血,提取核酸,用套式聚合酶链反应(PCR)扩增HIV-1的tat基因,并进行核酸测序,生成系统进化树。结果长期不进展者、无症状感染者及AIDS患者的tat基因序列与相应亚型的国际流行株序列比较,各组均存在与流行株序列不一致的氨基酸变异,而且某些变异具有组间特异性。在系统树分析中各组聚集在一起,呈混合分布。结论中国HIV/AIDS患者tat第一外显子一些位点的基因变异可能与疾病进展有关,某些位点的基因变异可能会降低病毒的复制能力。
Objective To study the characteristics of human immunodeficiency virus (HIV-1) tat exonl gene mutations among HIV-1 infected people in China.So as to find the ralationship between the tat gene mutation and the disease progression.Methods We extracted PBMC genome DNA from the whole blood samples of HIV-1 carriers and AIDS patients. The gene sequences of the Tat exonl were amplified by nest-polymerase chain reaction (nest-PCR). Products were sequenced directly,the sequences were aligned,translated and analyzed.Results There is difference in sequence variation between China HIV-1 in fected individuals and their according international ones. This difference is also exist among the LINP,asymptomatic HIV-1 infected individuals and AIDS patients.Individuals of three groups distribute together in the phelogenetic tree analysis.Conclusion some of the mutations of tat exonl could increase/decrease the ability of regulation transcription of Tat protein ,so as to increase/decrease the viral replication ability.
