摘要 目的观察急性高眼压时水通道蛋白-4(AQP4)在大鼠眼组织的表达变化,以探讨AQP4与某些眼压异常性疾病的关系。方法前房加压灌注法制作大鼠急性高眼压模型;应用免疫组化和原位杂交法观察AQP4及其mRNA在眼组织的表达变化,并进行半定量分析。结果与对照组相比,AQP4及其mRNA在急性高眼压大鼠视网膜上的表达明显增强,主要集中在节细胞层、内核层。结论急性高眼压状态下,AQP4及其mRNA在视网膜上的表达增加,提示AQP4可能参与了高眼压所致视网膜损伤的病理生理过程。 Objective To investigate the expressional changes of AQP4 and its mRNA under acute ocular hypertension(AOH) so as to study the role of AQP4 in some diseases associated with pressure alteration. Methods The animal model of AOH was made by perfuming physiological saline into the anterior chamber of rats;in situ hybridization and immunohistoehemistry were respectively used to evaluate AQP-4 and its mRNA expression under AOH ; then was quantitated by computerized morphometry. Resuits As compared with the control group, there appeared a significant increase in AQP-4 and its mRNA expression in the retina of experimental group, especially in the ganglion cell layer (GCL) and the inner nuclear (INL), Conclusion AQP4 may play an important role in the rat retinal injury under AOH.
