二氮嗪与1,6-二磷酸果糖伍用对大鼠心脏保存作用的初步研究 被引量:3

摘要 目的观察二氮嗪(DZ)与1,6-二磷酸果糖(FDP)伍用在心脏保存中的作用及量效关系。方法SD大鼠48只,随机分为6组,对照组(St.ThomasⅡ号液),D组(St.ThomasⅡ号液中含DZ100μmol/L),F组(St.ThomasⅡ号液中含FDP10mmol/L),DF组(分3个亚组,分别为St.ThomasⅡ号液中含DZ 25μmol/L与FDP 2.5 mmol/L、DZ 50μmol/L与FDP 5mmol/L、DZ100μmol/L与FDP 10mmol/L)。利用大鼠离体心脏工作模型,观察心脏在4℃条件下保存6h后心功能及冠脉流出液中乳酸脱氢酶(LDH)及磷酸肌酸激酶(CK)的变化。结果DZ、FDP单用或合用,复灌期心率、左室收缩压、左心室压力变化率、冠状动脉流出量均优于对照组,而左室舒张末压力以及冠脉流出液中CK及LDH的含量则明显低于对照组。其中DZ50μmol/L与FDP 5mmol/L、DZ100μmol/L与FDP 10mmol/L的结果显著优于其他各组,二者相比差异无统计学意义。结论DZ、FDP单用或合用均能提高St.ThomasⅡ号液心肌保存效果,两药合用明显优于单药。 Objective To investigate the effects of diazoxide (DZ) in combination with fructose 1,6 diphosphate (FDP) on heart preservation in rats. Mathods Forty-eight SD rats were randomly divided into six groups;the control group(n=8,St. Thomas Ⅱ solution) ;the D group( n= 8 , St. Thomas Ⅱ solution with DZ100μmol/L) ;the F group(St. Thomas Ⅱsolution with FDP 10mmol/ L) ;DF1 group (n=8,St. Thomas Ⅱ solution with DZ25μmol/L and FDP 2.5mmol/L) ;DF2 group(n=8,St. Thomas Ⅱ solution with DZ50gmol/L and FDP 5.0mmol/L);DF3 group(n= 8, St. Thmoas Ⅱ solution with DZ100gmol/L and FDP 10mmol/L). Working model of isolated rat heart was used. The hearts were stored at 4℃ for 6h, then the cardiac contractile function and myocardial enzyme leakage in the coronary effluent were detected. Results After stored at 4℃ for 6h, in DZ or FDP used alone or in combination groups, the heart rate, left ventricular systolic pressure, the maximal change of intraventricular pressure rise or decline (±dp/dtmax) and coronary flow were significantly higher than that of control group(P〈0.05 or P〈0.01 ); and the left ventrieular end-diastolic pressure, the leakage of myocardial enzymes (lactate dehydrogenase and creatine kinase)was significant lower than that of control group(P〈0.05 or P〈0.01). Group DF2 and DF3 had better effects, but there were no significant changes between the two groups(P〉0.05). Conclusion DZ and FDP used in combination could enhance myocardial protection during long-time hypothermic heart preservation.
