黑林1号杨组培叶片不定根的发生与内、外源激素关系的研究 被引量:11

摘要 采用石蜡切片法观察了黑林1号杨(小黑杨×波兰15ACL)叶片在外源激素0.3 mg/L NAA的诱导下不定根的发生过程。结果表明,诱导10 d左右,叶脉处薄壁细胞开始旺盛活动并脱分化,形成不定根根原基,随后这些根原基细胞群不断分裂和分化,15 d以后组培叶片上陆续形成肉眼可观察到的不定根。采用间接酶联免疫法测定叶片生根过程中内源激素IAA、ZR、GA3和ABA的动态变化,探讨了内、外源激素与叶片不定根发生的关系。结果显示:不定根的发生与内源IAA水平状况有密切关系;ZR对不定根根原基的发生没有明显的作用,但在不定根的进一步生长发育阶段中发挥着重要作用;低水平的GA3、高水平ABA和较高比值的ABA/GA3利于不定根的发生。 Using paraffin mounting method, the development process of adventitious roots induced by 0.3 mg/L exogenous NAA in leaves of (P.simonii × P. nigra) × P.15 ACL was observed. The results show that adventitious root primordium is generated from parenchyma cells which vigorously activated and de - differentation in vein of (P. simonii × P. nigra ) × P.15 ACL leaves after 10 days. With the division and differentiation of root primordium cells, adventitious roots seen by naked eyes formed at last after 15 days. The kinetic changes of endogenous hormones IAA, ZR, GA3 and ABA during rooting of leaves was measured with ELISA. The relationship between root generation and endogenous and exogenous hormones were discussed. It reveals that: the level of endogenous IAA ties up with adventitious roots generation. ZR doesn't influence adventitious roots generation obviously, but plays an important role in adventitious roots growth for going step further. Low level of GA3, high level of ABA and high ratio of ABA/GA3 are useful to generate adventitious roots.
