摘要
目的探讨RASSF1A(RAS相关区域家族1A)基因转录表达与卵巢癌发生、发展的关系。方法2003年7月至2005年2月蚌埠医学院附属医院采用逆转录-聚合酶链反应(RT—PCR)的方法检测42例卵巢恶性肿瘤患者癌组织、腹腔冲洗液(或腹水)中RASSF1A mRNA的表达水平,以厦11例正常卵巢和19例卵巢良性肿瘤组织中RASSF1A mRNA的表达。结果RASSF1A mRNA在所有正常和卵巢良性肿瘤组织中均表达,而在卵巢恶性肿瘤组织中存在较高的表达缺失率(57.1%)。RASSF1A mRNA的表达缺失率与卵巢恶性肿瘤的临床分期有关,Ⅲ-Ⅳ期卵巢恶性肿瘤组织中RASSF1A mRNA的表达缺失率显著高于Ⅰ~Ⅱ期,淋巴结转移阳性者RASSF1A mRNA表达缺失率(73.9%)明显高于淋巴结转移阴性者(36.8%)(P〈0.05);腹腔冲洗液(或腹水)细胞学阳性者。其RASSF1A mRNA的表达缺失率显著升高。结论RASSF1A转录表达缺失可能参与调控卵巢癌的发生、发展过程,并且提示RASSF1A表达可作为判断卵巢癌预后的一个指标。
Objective To investigate the relationship between the transcriptional expression of RASSF1A ( Ras association domain family 1A gene) and oncogenesis, development of ovarian cancer. Methods RT- PCR was used to detect the expression of RASSF1A mRNA in 42 malignant ovarian tumor tissues and aseites (or peritoneal fluid), 11 normal ovarian tissues and 19 benign ovarian tumor tissues in the Affiliated Hospital of Bangbu Medical College from July 2003 to February 2005. Results The RASSF1A mRNA was identified in all noneaneer tissues but not found in 57.1% malignant ovarian tumor tissues. Much more frequent in advanced tumor tissues, loss of RASSF1A mRNA was correlated with stage ( loss rate of RASSF1A mRNA was positively correlated with clinical stage). The rate of loss of RASSF1A mRNA in Ⅲ - 1V was more higher than Ⅰ-Ⅱ (loss rate of RASSF1A mRNA was higher in stage Ⅲ-Ⅳ than in stage Ⅰ-Ⅱ ). The loss rate of RASSF1A mRNA was significantly higher in the patients with positive lymph node metastasis (73.9%) than in those without lymph node metastasis (36.8%) (P 〈 0. 05) . The loss rate of RASSF1A mRNA was significantly higher in the patients with positive ascites or peritoneal liquid. Conclusion Loss or abnormal down - regulation of RASSF1A mRNA is closely related to oncogenesis and development of ovarian cancer. It may be a prognostic marker of ovarian cancer.
