摘要 目的探讨氧疗对缺血缺氧家兔炎症细胞因子TNF-α和IL-6 mRNA表达的影响。方法将15只家兔随机分为实验组与对照组,放入模拟海拔4000 m高原低压舱内,复制失血性休克模型。对照组只给予扩容治疗,实验组在扩容的同时给予氧疗,于不同时相点进行指标检测。结果休克时,外周血WBC中TNF-α和IL-6 mRNA表达均增高,分别于治疗后14、h达高峰,但实验组表达增加的幅度小于对照组,与血浆内毒素(LPS)水平及丙二醛(MDA)含量变化相似;同时实验组的肠黏膜损伤程度轻于对照组。结论缺血缺氧后体内炎症细胞因子表达增加,氧疗可以使TNF-α和IL-6 mRNA的表达降低,有利于减轻全身炎症反应和组织器官损害。 Objective To investigate the effects of oxygen therapy an the expression of cytokines TNF - α and 1L - 6mRNA in the rabbits with ischemia and hypoxia. Methods Fifteen rabbits were exposed to a simulated 4000 m high altitude in a hypobaric chamber and at random divided into two groups: trial and control. Hemorrhagic shock was induced by bleeding. After 1 hour, the rabbits were treated with volume expression. In addition, the trial group was administered oxygen therapy. Results After shock, the expression of TNF - α and IL - 6mRNA in WBC increascd, peaked at 1h and 4h after treatment, but the range of changes in the triMgroup was less than that of the control, which was similar to the changes of LPS level and MDA concentration in plasma. The intestinal mucosa injury of the trial was lighter than that of the control. Conclusions The expressions of imflammatory cytokines up - regulates during ischemia and hypoxia. Oxygen therapy eontributes to down - regulation of the expressions of TNF- α and IL- 6mRNA, and systemic inflammatory reaction may be relieved.
