用人角膜给猴移植的实验研究

摘要 用人角膜给猕猴移植,共7只。第2、3、6号猴供片直径是7.5mm,植床直径是7.0mm;第1、4、5、7号猴的供片直径力6.2mm,植床直径为6.0mm;结果:第2、3只猴植片透明达1年以上,第6只猴已透明20个月,现仍在观察中,第1只猴17天死亡,死前植片透明,第4、5、6只猴,植片透明4.5~8个月,小植片比大植片透明时间较长。实验猴№2术后8个月时,植片内皮细胞的密度为2570个/mm^2,术后13个月时为1800个/mm^2,植片内皮的丢失率超过生理下降速度。扫描电镜检查证实,人角膜内皮可以在猴眼上生存一个相当长的时间。免疫排斥反应,早期和轻型者,可用激素控制;后期和新生血管明显者,激素难以奏效。这一实验模型的建立,为今后研究异种角膜移植提供了有用的材料。 Human corneal grafts were transplanted into 7 rhesus monkey corneas. 4 eyes (monkeys No. 1, 4, 5, and 7) received a 7.5 mm donor in a 7.0mm recipient bed seperately and 3 others (No. 2, 3 and 6) received a 6.2 mm donor in a 6. Omm recipient hed by means of continuous 10-0 nylon sutures。The results showed that the graft of monkey No. 1 was transparent, but it died 17 days after operatron. The grafts of the recipient monkeys (No. 2, 3 and 6) remained transpatent for 12 months. The graft of recipient monkey No. 4, 5 and 7 had been transplant for 4-8 months. Examinations by specular microscopy showed that the endothelium density of monkey No. 4, 5 and 7 were 1666-2000 cells/mm 3.5-4 months after operation. The endothelium density of monkey No. 2 was 2570 cells/mm 8 months after operation and was reduced to 1800 cells/ mm 13 months after operation. The decrement rate of graft endothelium was higher than physiological. Data ohtained by SEM showed that human corneal endothelium could survive in monkey's cornea for quite long time. The graft rejection in early or mild cases could be controlled, but it would not work in advanced or severe cases or with evident vascularization. The preliminary experience of the animal models furnished us valuable data for further study of heterografting of cornea.
