摘要
RNA干扰(RNAi)是由双链RNA(dsRNA)介导的、序列特异地引起转录后基因沉默的现象,是生物体内抵御转座子和病毒感染的重要保护机制之一,也是在生长发育过程中调节内源基因表达的重要途径。RNAi可作为一种简单有效的代替基因剔除的工具应用于功能基因组学和疾病的基因治疗研究。现综述RNAi机制、作用特征及RNAi相关技术在小鼠卵母细胞和胚胎发育研究中的应用。
The phenomenon of RNA interference ( RNAi ) is a response to double-stranded RNA (dsRNA), which results in posttranscriptional sequence-specific gene silence. RNAi and related mechanisms are an essential part of a complex defense mechanism against transposable elements and viruses. As a comphment of gene knockout, RNAi works as a simple, effective tool for promoting the study of functional genomies and gene therapy. In this view,the mechanism and character of RNAi as well as the applicaton in the study of development of mouse ooeytes and embryos is discussed.
